Colon (2-1) allowed one run on seven hits in winning his second straight start to give New York its first victory in the four-game series. He struck out six and walked one to lower his ERA to 2.88.
Cano provided Colon with the only runs he needed with his sixth homer, a first-inning blast off Mark Buehrle (1-3), who fell to 1-8 lifetime against the Yankees.
Mariano Rivera, who had blown his last two save opportunities, threw a perfect ninth for his eighth save.
Buehrle pitched well after Cano's homer, going seven innings and permitting three runs on six hits. He fanned five and walked two.
Carlos Quentin went 3-for-3 and Adam Dunn had a pair of hits, including an RBI single in the sixth, for Chicago.
